    Not really good advice is it, because the OP has already admitted that they have no management experience. They have been thrown in at the deep end by the owner/top management in a sink or swim situation.
    If I were the OP, then I would call a meeting with all the staff under my control, and tell then to stop moaning and get on with their jobs - make eye contact with everyone when they say that, and let them know that there are grievance procedures if they are unhappy, and to use them. Stress that they can work as a team, or changes will have to be made, making sure they know that no staff member is indispensable.
    If the cranky/moany person still keeps on behind the backs of others, then they will have to be told to stop, or possibly face a verbal warning.

    UPDATE
    Cranky has done the obvious and put a sick note in for a month, I was expecting that.
    She always throws a tantrum then goes off sick, burying her head in the sand and hoping it will all go away!

    Then it's very important that while she's off you ask your manager about suitable training, maybe shadowing, on return to work interviews, and make sure you know your company's policy on sick pay, whether she needs to see Occupational Health etc.

    Excessive sick leave? Possible grounds for a fair (as opposed to unfair) dismissal
