NHS Job Offer conditional on references...

Why would you be asked to hand your notice in, simply because you were looking for another job? And why do you think you 'will sort of have to' resign straight away?
What sort of job is it you do that they'd expect you to resign because you've applied for another job? I can't think of any occupation which would fall into that category.
I work in an SME with a particularly hostile working culture, and a lot of people in this position take job hunting as a partner would take cheating. I basically have to "out" myself as job-hunting at the point the reference is asked, which means at best I will be sidelined at work politically (which will make day to day working very difficult as obtaining resource to get my job done will be much harder, which could lead to negative performance reviews) and at worst I could be informally asked to resign.
In the public sector and bigger companies this is probably seen as normal, but companies with <50 people especially if they've spent time training me probably less so.0 -

If you're going to leave then does anything that I've highlighted matter?

It's more that as soon as the reference is asked for, I'll be asked to hand my notice in, and on good faith, I will sort of have to as I don't want to burn bridges.
Personally I wouldn't be handing my notice in until all the references, DBS etc were through and there was no chance of the job offer being rescinded. Good faith doesn't come into it.All shall be well, and all shall be well, and all manner of things shall be well.
Pedant alert - it's could have, not could of.0
