When to transfer money

Hello
I want to pay extra on my mortgage but I can only pay £500 per calender month. I have a savings account and mortgage with Nationwide, Nationwide say that paying from my savings account to my mortgage will take two days so my question is - do I wait until the 1st May to transfer the money or can I do it now?
