I live away from home 5 nights per week and pay council tax with 25% discount.
can my wife also claim 25% discount on house where i spend 2 nights per week?

No, the property with your wife to which you return is also 'sole or main ' residence therefore she cant claim a discount.
You can however claim a second home discount on the home that you live in during the week whilst your away , you shouldn't be in receipt of a 25% single person discount on the property you live in during the week.I no longer work in Council Tax Recovery but instead work as a specialist Council Tax paralegal assisting landlords and Council Tax payers with council tax disputes and valuation tribunals. My views are my own reading of the law and you should always check with the local authority in question.0 -

However, why can't my wife and I be regestered at different properties for our main residence each as sole occupants if : we have separate bank accounts at different addresses, on electoral role at different addresses, and previously (genuinely) been living apart for several years etc?0 -
I see where you are coming from.
If you count the other place as your main residence and have sufficient documentation to prove it (bank accounts, electoral role) then you may well both be able to claim single resident status at each of your properties.
I'd take further advice on the matter though, but you could well be right.0 -

Because, as I understand the situation, you still live together as man and wife in your main residence and the intention is that you will do so in the future and your 5 day a week home is only temporary.If you are querying your Council Tax band would you please state whether you are in England, Scotland or Wales0
